چکیده

In the treatment of influenza, Neuraminidase inhibitors (NAIs) (Oseltamivir and Zanamivir) play a major role. The emergence variants influenza A (H1N1) pdm09 virus resistant to Oseltamivir is matter great concern as it limits its usage. Therefore, vigilant monitoring for Oseltamivir-resistant viruses has been recommended by World Health Organization (WHO). Our study aimed screen NAI drug resistance during outbreak 2015-16 in North-Western India. total 640 H1N1pdm09 virus-positive samples were screened WHO allelic discrimination real-time RT-PCR protocol. PCR protocol can detect presence single nucleotide polymorphisms (SNPs), H275Y mutation detected this method which causes Oseltamivir. Sanger sequencing partial fragment NA gene (fragment IV), 90 performed confirm NA-H275Y mutation. susceptibility 20 randomly selected isolates was tested using inhibition chemiluminiscence based assay. Among positive tested, one sample (0.15%) confirmed also. All star assay found be sensitive an easy, rapid high-throughput detection Systematic regular surveillance Influenza essential monitor spread drug-resistant strains.

I223R Mutation in Influenza A(H1N1)pdm09 Neuraminidase Confers Reduced Susceptibility to Oseltamivir and Zanamivir and Enhanced Resistance with H275Y

BACKGROUND Resistance of pandemic A(H1N1)2009 (H1N1pdm09) virus to neuraminidase inhibitors (NAIs) has remained limited. A new mutation I223R in the neuraminidase (NA) of H1N1pdm09 virus has been reported along with H275Y in immunocompromised patients. The aim of this study was to determine the impact of I223R on oseltamivir and zanamivir susceptibility. METHODS The NA enzymatic characteristi...

Characteristics of a Widespread Community Cluster of H275Y Oseltamivir-Resistant A(H1N1)pdm09 Influenza in Australia

BACKGROUND Oseltamivir resistance in A(H1N1)pdm09 influenza is rare, particularly in untreated community cases. Sustained community transmission has not previously been reported. METHODS Influenza specimens from the Asia-Pacific region were collected through sentinel surveillance, hospital, and general practitioner networks. Clinical and epidemiological information was collected on patients i...

Oseltamivir-resistant influenza A(H1N1)pdm09 virus in southern Brazil.

The neuraminidase (NA) genes of A(H1N1)pdm09 influenza virus isolates from 306 infected patients were analysed. The circulation of oseltamivir-resistant viruses in Brazil has not been reported previously. Clinical samples were collected in the state of Rio Grande do Sul (RS) from 2009-2011 and two NA inhibitor-resistant mutants were identified, one in 2009 (H275Y) and the other in 2011 (S247N)....

Determination of Oseltamivir Resistance Level by an H275Y Genotyping Assay among Influenza A (H1N1) Viruses in Hamadan Province, Iran

Introduction: Epidemics and deaths caused by influenza viruses are an important concern worldwide. The use of neuraminidase inhibitors such as oseltamivir is an effective and valuable way to treat the diseases caused by these viruses. However, the mutation in several parts of the gene leads to the emergence of drug-resistant strains, and an ever-increasing rise in drug-resistant strains is a gl...
