Detection of H275Y Mutation Conferring Oseltamivir Drug Resistance in Influenza A (H1N1) pdm09 Virus

In the treatment of influenza, Neuraminidase inhibitors (NAIs) (Oseltamivir and Zanamivir) play a major role. The emergence variants influenza A (H1N1) pdm09 virus resistant to Oseltamivir is matter great concern as it limits its usage. Therefore, vigilant monitoring for Oseltamivir-resistant viruses has been recommended by World Health Organization (WHO). Our study aimed screen NAI drug resistance during outbreak 2015-16 in North-Western India. total 640 H1N1pdm09 virus-positive samples were screened WHO allelic discrimination real-time RT-PCR protocol. PCR protocol can detect presence single nucleotide polymorphisms (SNPs), H275Y mutation detected this method which causes Oseltamivir. Sanger sequencing partial fragment NA gene (fragment IV), 90 performed confirm NA-H275Y mutation. susceptibility 20 randomly selected isolates was tested using inhibition chemiluminiscence based assay. Among positive tested, one sample (0.15%) confirmed also.
